What’s right: First Male Pill with No Side Effects Clears Safety Test

A major breakthrough in reproductive health: the first-ever non-hormonal male birth control pill, YCT‑529, has passed its initial human safety trial—a giant leap toward a more balanced future in family planning. In a Phase 1a study of 16 healthy men (all having had vasectomies to preserve fertility), doses up to 180 mg were found to be well-tolerated, with no effects on testosterone, mood, heart rate, libido, or inflammatory markers.

Unlike hormone-based contraceptives, YCT‑529 works by blocking the retinoic acid receptor alpha (RAR‑α) in the testes, which halts sperm production while leaving hormone levels untouched. In animal studies, it achieved 99% effectiveness in preventing pregnancy, with fertility fully restored within weeks of stopping the drug.

The trial's results have paved the way for ongoing 28‑ and 90‑day studies in more participants, confirming safety and measuring sperm reduction efficacy. If trials continue to succeed, YCT‑529 could reach the market by the end of the decade as the first reversible male contraceptive pill.

This development promises greater reproductive equity, giving men a direct option to share contraceptive responsibility—without unwanted side effects.
